Minimal Media Agar Plates are a defined agar medium for cultivation and characterization of E. coli and other bacteria under nutrient-limited conditions. Glucose serves as the primary carbon source, ammonium provides inorganic nitrogen, and phosphate maintains buffering capacity while supplying phosphorus. Magnesium and calcium supply divalent cations required for cellular processes, and thiamine provides vitamin B1.

These plates are suitable for selective growth based on metabolic requirements, phenotypic screening of auxotrophs and prototrophs, and maintenance of strains using appropriate antibiotic supplements when required. Prepared as ready-to-use agar plates for routine microbiological workflows.

  • 1mM Calcium Chloride
  • 42.2mM Sodium Phosphate Dibasic
  • 22mM Potassium Phosphate Monobasic
  • 18.7mM Ammonium Chloride
  • 0.1% Glucose

  • Auxotrophy/Prototrophy Screening: Identify nutrient requirements and select prototrophic revertants of Escherichia coli and other minimal medium-competent strains.

  • Auxotrophic Complementation Selection: Maintain/select plasmids or genomic integrations that complement metabolic defects in Escherichia coli on defined glucose minimal agar.

  • Carbon Source Utilization Assays: Assess growth and metabolism under defined conditions (glucose-based), or with alternative carbon sources, in strains such as Escherichia coli and Pseudomonas putida.

  • Metabolic and Gene Knockout Phenotyping: Characterize effects of pathway deletions on growth and fitness in defined media.

  • Colony Isolation and CFU Enumeration: Obtain isolated colonies and quantify viable counts under controlled, minimal conditions.

  • Adaptive Laboratory Evolution (ALE): Evolve strains like Escherichia coli or Salmonella enterica under defined nutrient limitations.

  • Antibiotic Susceptibility in Defined Media: Evaluate antimicrobial effects without confounding rich-medium components.

  • Catabolite Repression and Diauxic Growth Studies: Probe regulatory responses to defined glucose availability.
