Mueller Hinton Agar Plates are ready-to-use, non-selective agar plates suitable for the cultivation of a broad range of aerobic, non-fastidious bacteria, including Escherichia coli, Staphylococcus aureus, Pseudomonas aeruginosa, and Enterococcus faecalis. The medium provides a uniform surface appropriate for antimicrobial disk diffusion testing and general microbiology workflows.

Applications include routine cultivation of clinical and environmental isolates, antimicrobial susceptibility testing by disk diffusion following standardized methods, and maintenance of reference strains.

Store plates at 2–8 °C in the dark. Allow plates to reach room temperature before inoculation. Incubate aerobically at 35–37 °C as appropriate for the organism and method.

  • 3.8% Mueller Hinton Agar
  • Kirby–Bauer Disk Diffusion AST: Standardized 3.8% Mueller–Hinton Agar supports reliable antibiotic zone-of-inhibition testing.

  • MIC by Gradient Diffusion Strips: Used to determine minimum inhibitory concentrations with Etest/gradient strips.

  • Quality Control with Reference Strains: Routine QC of antibiotic performance using strains such as Escherichia coli ATCC 25922, Staphylococcus aureus ATCC 25923, and Pseudomonas aeruginosa ATCC 27853.

  • Nonfastidious Bacterial Cultivation: Growth and maintenance of aerobic and facultative organisms for downstream testing.

  • AST of Fastidious Organisms (Supplemented): When supplemented with 5% sheep blood and β-NAD, suitable for testing Streptococcus pneumoniae and Haemophilus influenzae.

  • Trimethoprim/Sulfonamide Testing: Low thymidine/thymine content enables accurate susceptibility testing for these agents in Enterobacterales and other pathogens.

  • Education and Training: Demonstrations of antibiotic susceptibility and resistance profiling in teaching labs.

  • Antimicrobial Diffusion Studies: Evaluation of diffusion characteristics and pigment production with organisms like Pseudomonas aeruginosa.
