Mueller Hinton II Cation Adjusted Agar Plates are a defined, nonselective medium used for antimicrobial susceptibility testing by standardized disk diffusion and dilution methods. The cation-adjusted formulation provides specified levels of calcium and magnesium appropriate for these applications and supports the cultivation of nonfastidious aerobic and facultative bacteria.

The medium is routinely used with organisms such as Staphylococcus aureus, Enterococcus faecalis, Escherichia coli, Klebsiella pneumoniae, Pseudomonas aeruginosa, and Salmonella enterica, and is compatible with a broad range of antimicrobial agents prepared according to established guidelines.

  • 1.75% Casamino Acids
  • 0.30% Beef Extract
  • 0.15% Soluble Starch
  • 1.50% Agar
  • 20µg/mL Calcium Chloride Dihydrate
  • 10µg/mL Magnesium Chloride Hexahydrate
  • Disk Diffusion Antimicrobial Susceptibility Testing (Kirby-Bauer): Standardized cation-adjusted medium for reliable zone diameters with nonfastidious aerobes such as Staphylococcus aureus, Escherichia coli, and Pseudomonas aeruginosa.

  • Gradient Diffusion MIC (Etest) Determination: Used to determine minimum inhibitory concentrations for clinical isolates on a reproducible, cation-controlled surface.

  • Antibiotic Potency/Bioassay Testing: Employed in assay systems to quantify antimicrobial activity and lot-to-lot consistency of drug products.

  • Quality Control of Susceptibility Testing: Verification with control strains (e.g., Escherichia coli ATCC 25922, Staphylococcus aureus ATCC 25923, Pseudomonas aeruginosa ATCC 27853) to confirm media and disc performance.

  • Routine Isolation and Cultivation of Nonfastidious Bacteria: Growth and enumeration of organisms like Enterococcus faecalis, Klebsiella pneumoniae, and Acinetobacter baumannii.

  • Beta-lactam Resistance Screening: Cefoxitin/oxacillin disc screening for MRSA detection in Staphylococcus aureus on a standardized agar matrix.

  • Streptococcal Susceptibility Testing (Supplemented): Use with 5% sheep blood for fastidious organisms such as Streptococcus pneumoniae to support reliable AST.

  • Haemophilus and Other Fastidious AST (Appropriately Supplemented): Use with required X/V factors or specialized supplements for organisms like Haemophilus influenzae when indicated by guidelines.

  • Verification of Antibiotic Disc Potency and Storage Stability: Periodic checks of disc performance using standardized lawns on cation-adjusted plates.
