LB Lennox Top Agar is a semi-solid overlay medium used for bacteriophage plaque assays and colony screening with E. coli host strains. The formulation is based on LB Lennox nutrients and a reduced agar concentration to facilitate diffusion and plaque formation.

Tryptone and yeast extract supply peptides, amino acids, and growth factors for E. coli, sodium chloride maintains osmotic balance, and agar provides a soft gel matrix suitable for overlays on LB base plates.

  • 1.00% Tryptone
  • 0.50% Yeast Extract
  • 0.50% Sodium Chloride
  • 0.75% Agar

  • Bacteriophage Plaque Assays: Soft agar overlays (0.75% agar) to titer and isolate phages on Escherichia coli lawns.

  • Phage Propagation and Enrichment: Overlay method to amplify phage stocks on susceptible hosts such as Escherichia coli and Salmonella enterica.

  • Generalized Transduction: P1 or P22 transduction workflows using top agar overlays for donor phage and recipient cells.

  • Plaque Purification: Serial plaque picking from soft agar overlays to obtain clonal phage isolates.

  • Host Range Testing: Overlay indicator lawns of different bacteria (e.g., Escherichia coli, Salmonella enterica) to assess phage infectivity.

  • Lambda Phage Screening: Library screening and recombinant phage detection via plaque formation on Escherichia coli.

  • Lysogeny and Immunity Tests: Assess lysogen formation and superinfection immunity by plaque morphology in soft agar.

  • Phage-Resistance Selection: Identify and recover bacterial mutants that resist infection within overlay lawns.
