LB Top Agar is a semi-solid agar medium designed for overlay applications in bacteriophage plaque assays and bacterial screening. It provides a nutrient base suitable for the growth of E. coli strains and forms a soft gel matrix appropriate for top-layer pours over solidified base plates.

This medium is commonly used with a pre-poured LB or similar basal agar to facilitate plaque visualization and colony isolation. It can be supplemented with antibiotics or other additives according to experimental requirements and is typically melted and tempered prior to overlay.

  • 1.00% Tryptone
  • 0.50% Yeast Extract
  • 1.00% Sodium Chloride
  • 0.75% Agar
  • Bacteriophage Plaque Assays: Soft-agar overlays to visualize and quantify phage plaques on lawns of Escherichia coli, Salmonella, or Pseudomonas.

  • Phage Titration (PFU Determination): Accurate enumeration of infectious phage particles using uniform bacterial overlays.

  • Plaque Purification: Serial picking and re-plating of individual plaques to isolate clonal phage populations.

  • Host Range and EOP Testing: Comparative efficiency-of-plating assays across multiple bacterial hosts (e.g., E. coli, Salmonella).

  • Bacteriophage Transduction Assays: Soft-agar overlays for generalized/specialized transduction workflows (e.g., P1 in E. coli).

  • Bacteriocin and Antimicrobial Spot Tests: Indicator lawns to assess inhibition zones from bacteriocins, antibiotics, or phage-derived lysins.

  • Phage Lysogeny and Induction Studies: Monitoring lysogen formation and prophage induction via plaque morphology changes on bacterial lawns.
