LB Luria Broth is a nutrient medium for the cultivation of non-fastidious bacteria, including E. coli. It is commonly used for routine propagation, maintenance of laboratory strains, and preparation of starter cultures.

This formulation features reduced sodium chloride content (low-salt LB), providing a lower ionic strength medium compared with standard LB variants.

  • 1.00% Tryptone
  • 0.50% Yeast Extract
  • 0.05% Sodium Chloride

  • Routine Cultivation of Escherichia coli: Growth and maintenance of recombinant and laboratory E. coli strains in a low-salt (0.05% NaCl) LB medium.

  • Plasmid Cloning and Propagation: Amplification of plasmids with antibiotic selection in liquid culture for minipreps/midipreps.

  • Starter Cultures for Protein Expression: Inoculation and overnight growth of Escherichia coli pre-cultures prior to induction in richer media.

  • Transformation Recovery/Outgrowth: Post heat-shock or electroporation recovery of Escherichia coli in a gentle, low-salt medium.

  • Colony Picking and Glycerol Stocks: Expansion of single colonies to prepare archival glycerol stocks and working cultures.

  • Phage-Related Work: Low-salt conditions suitable for certain bacteriophage propagation and assays with Escherichia coli hosts.

  • General Bacterial Growth: Culturing non-fastidious bacteria such as Bacillus subtilis under conditions where reduced NaCl is preferred.

  • pH: 7.10 ± 0.10
