LB Luria Broth is a nutritionally balanced liquid medium for routine cultivation and maintenance of enteric bacteria, including E. coli, in general cloning and plasmid propagation workflows. It supplies peptides, amino acids, vitamins, and minerals suitable for overnight cultures, starter inocula, and strain propagation in shaking or static conditions.

The medium serves as a base for antibiotic supplementation and carbohydrate additions when required for selection or experimental design. It is commonly used for propagation of plasmid-bearing and non-plasmid-bearing E. coli strains, preparation of glycerol stocks, and routine microbial quality control.

  • 1.00% Tryptone
  • 0.50% Yeast Extract
  • 0.05% Sodium Chloride
  • Bacterial Cultivation: Routine growth of Escherichia coli in liquid culture for general microbiology and molecular biology workflows.

  • Cloning and Plasmid Amplification: Culturing recombinant Escherichia coli for plasmid DNA preparation (mini-/midi-/maxi-preps).

  • Antibiotic Selection and Plasmid Maintenance: Growth of transformed Escherichia coli with appropriate antibiotics to maintain selectable markers.

  • Transformation Recovery/Outgrowth: Post-transformation recovery of Escherichia coli prior to plating or downstream applications.

  • Starter Cultures for Protein Expression: Preparing overnight cultures of recombinant Escherichia coli to inoculate expression media.

  • Glycerol Stock Preparation: Growing Escherichia coli prior to cryopreservation for strain and plasmid archiving.

  • Phage Work: Propagation of filamentous or lytic phages (e.g., M13, T7) in susceptible Escherichia coli hosts.

  • Growth Curves and OD600 Monitoring: Assessing Escherichia coli growth kinetics and media performance.

  • General Enterobacteria Culture: Non-fastidious growth of related bacteria such as Salmonella enterica for basic studies.

  • pH: 7.10 ± 0.10
