LB Broth is a standard nutrient medium containing tryptone, yeast extract, and sodium chloride. It is used for routine cultivation and maintenance of E. coli and related enteric bacteria in liquid culture. The formulation provides amino acids, peptides, vitamins, and ionic strength suitable for general growth and propagation, including preparation of overnight cultures and plasmid propagation when selective agents are added separately.

  • 1.0% Tryptone
  • 0.5% Yeast Extract
  • 1.0% Sodium Chloride
  • Routine Cultivation of Bacteria: General growth and maintenance of non-fastidious microbes, especially E. coli, using a nutritionally balanced 1.0% Tryptone, 0.5% Yeast Extract, 1.0% Sodium Chloride formulation.

  • Plasmid Propagation: Culturing transformed E. coli for plasmid amplification and downstream DNA purification.

  • Protein Expression: Growing recombinant E. coli cultures for inducible protein production.

  • Antibiotic Selection in Liquid Culture: Maintaining selection pressure for plasmid-bearing E. coli by supplementing with appropriate antibiotics.

  • Overnight and Starter Cultures: Preparing inocula for scale-up, minipreps, analytical assays, and glycerol stock creation.

  • Transformation Recovery: Post-heat shock or electroporation outgrowth of E. coli prior to selection.

  • Growth Curve and OD600 Assays: Monitoring bacterial kinetics and fitness in standardized LB conditions.

  • Bacteriophage Workflows: Culturing host E. coli for propagation of filamentous and lytic phage in liquid media.

  • pH: 7.00 ± 0.10
