LB Agar Plates are solid LB medium used for routine cultivation and maintenance of E. coli and other nonfastidious bacteria. The formulation provides peptides and amino acids from tryptone and vitamins from yeast extract, with sodium chloride to maintain osmotic balance and agar as the solidifying agent. Used for colony isolation, propagation, and cloning workflows that do not require antibiotic selection.

  • 1.0% Tryptone
  • 0.5% Yeast Extract
  • 1.0% Sodium Chloride
  • 1.5% Agar
  • Bacterial Culture and Colony Isolation: Growth and isolation of single colonies from mixed or pure cultures of non-fastidious bacteria such as E. coli and Bacillus subtilis.

  • Plasmid Transformation Selection: Selection of recombinant E. coli transformants when supplemented with appropriate antibiotics.

  • Colony-Forming Unit (CFU) Enumeration: Quantification of viable bacteria by serial dilution and plating.

  • Cloning and Screening: Blue/white screening and colony PCR workflows when supplemented with X-gal/IPTG and suitable vectors.

  • Strain Maintenance and Storage: Short-term storage and routine propagation of laboratory strains on plates.

  • Phage Work: Preparation of bacterial lawns (e.g., E. coli) for coliphage plaque assays using soft agar overlays.

  • Antibiotic Resistance Screening: Assessment of selectable markers or resistance phenotypes on supplemented plates.

  • Conjugation and Mating Assays: Selection of transconjugants in bacterial mating experiments (e.g., E. coli donor/recipient strains) with selective supplements.

  • Environmental Isolate Culturing: Initial culturing and colony picking from environmental or microbiome samples of non-fastidious bacteria.

  • Quality Control Checks: Monitoring for contamination of media, reagents, and workflows via surface streaking and plating.
