LB Agar Plates are a nutrient agar medium for routine cultivation and plating of E. coli and other nonfastidious bacteria. The solid medium supports colony isolation, maintenance, and propagation, and can be used as a base for selection when supplemented with appropriate antibiotics.

  • 1.0% Tryptone
  • 0.5% Yeast Extract
  • 1.0% Sodium Chloride
  • 1.5% Agar
  • Isolation of Single Colonies: Streaking to obtain clonal isolates of Escherichia coli and other nonfastidious bacteria.

  • Antibiotic Selection of Transformants: Plating transformed E. coli on LB agar supplemented with antibiotics to select plasmid-bearing colonies.

  • CFU Enumeration: Spread or pour plating for colony-forming unit counts from liquid cultures or environmental samples.

  • Maintenance of Bacterial Strains: Routine culture and short-term storage of lab strains (e.g., E. coli, Bacillus subtilis).

  • Cloning Workflows: Recovery and screening of colonies after ligation, Gibson assembly, or Golden Gate cloning.

  • Blue-White Screening: Screening of recombinant clones on LB agar with X-gal/IPTG and appropriate antibiotic.

  • High-Density Library Screening: Large-format plating (245 mm) for colony libraries and automated picking.

  • Replica Plating: Transferring colonies to selective/nonselective media to assess phenotypes or auxotrophies.

  • Phage Plaque Assays: Preparing bacterial lawns (e.g., E. coli) on LB agar with soft agar overlays for bacteriophage titration.

  • Conjugation and Mating Assays: Selection of transconjugants after bacterial conjugation on selective LB agar.
