LB Agar Plates are a nutrient agar medium for the cultivation and plating of laboratory strains of E. coli. The solid medium provides a surface suitable for streaking, colony isolation, and enumeration in routine microbiology workflows.

Common applications include growth and maintenance of recombinant E. coli, screening of transformants when combined with appropriate selective agents, and preparation of single colonies for downstream molecular biology procedures.

  • 1.0% Tryptone
  • 0.5% Yeast Extract
  • 1.0% Sodium Chloride
  • 1.5% Agar

  • Clonal Isolation and Streaking: Isolation of single colonies from mixed cultures of Escherichia coli and other lab strains.

  • Antibiotic Selection: Selection of plasmid-bearing transformants on plates supplemented with antibiotics.

  • Colony Screening: Blue-white screening and colony morphology assessment for recombinant clones.

  • Colony PCR and Downstream Workflows: Picking colonies for colony PCR, Sanger sequencing, and plasmid miniprep.

  • CFU Enumeration and Viability: Spread-plate or drop-plate counting of colony-forming units.

  • Transformation Recovery and Selection: Post-heat-shock or electroporation plating of Escherichia coli transformants.

  • Phage Workflows: Base layer for soft-agar overlay plaque assays with coliphages infecting Escherichia coli.

  • Strain Maintenance: Short-term maintenance, replica plating, and storage of working stocks.
