LB Agar Plates with Ampicillin-100, X-Gal, and IPTG are an LB agar medium for growth of ampicillin-resistant E. coli and visual screening of recombinant clones.

Ampicillin enables selection of plasmid-bearing transformants, X-Gal functions as a chromogenic substrate for β‑galactosidase, and IPTG induces lac operon expression, facilitating blue–white differentiation via α‑complementation in compatible host–vector systems.

  • 1.0% Tryptone
  • 0.5% Yeast Extract
  • 1.0% Sodium Chloride
  • 1.5% Agar
  • 100µg/mL Ampicillin Sodium Salt
  • 60µg/mL X-Gal
  • 0.1mM Isopropyl-B-D-thiogalactopyranoside(IPTG)
  • Blue-White Colony Screening: lacZ α-complementation on ampicillin selection to distinguish recombinant (white) from non-recombinant (blue) colonies in Escherichia coli.

  • Ampicillin-Resistant Transformant Selection: Isolation of successful transformants after ligation, Gibson Assembly, or Golden Gate cloning in E. coli.

  • Plasmid Maintenance and Propagation: Stable growth of AmpR plasmids in cloning strains of E. coli (e.g., DH5α, TOP10, JM109).

  • Colony Screening Workflows: Picking colonies for colony PCR, restriction analysis, and Sanger sequencing preparation.

  • Library Screening: Identification of inserts in cDNA/genomic libraries via lacZ disruption (white colony selection) in E. coli.
