LB Agar Plates with Animal-Free Soytone and Chloramphenicol-25 is a nutrient agar medium that supports the growth and selection of chloramphenicol-resistant E. coli strains. Animal-free soytone and yeast extract provide peptides, amino acids, vitamins, and growth factors; sodium chloride maintains osmotic balance; agar supplies a solid growth surface; and 25 µg/mL chloramphenicol enables selection of plasmid-bearing transformants.

  • 1.7% Animal-Free Soytone
  • 0.5% Yeast Extract
  • 1.0% Sodium Chloride
  • 1.5% Agar
  • 25µg/mL Chloramphenicol

  • Antibiotic Selection of Transformants: Selects and maintains chloramphenicol-resistant Escherichia coli colonies after transformation.

  • Plasmid Cloning and Maintenance: Propagation of CmR plasmids (cat) in Escherichia coli, ensuring plasmid retention during growth.

  • Colony Isolation and Screening: Streaking for single colonies, replica plating, and colony PCR of chloramphenicol-resistant transformants.

  • Bacterial Artificial Chromosomes (BACs): Selection and maintenance of chloramphenicol-marked BACs/cosmids/fosmids in Escherichia coli.

  • Dual-Plasmid Co-selection: Maintaining strains carrying multiple plasmids (e.g., pACYC/p15A origin) using chloramphenicol alongside other antibiotics.

  • Expression Strain Support: Culture of expression hosts harboring chloramphenicol-resistant helper plasmids (e.g., T7 lysozyme plasmids) in Escherichia coli.

  • CFU Enumeration Under Selection: Quantifying viable chloramphenicol-resistant cells via spread/plating assays.

  • Strain Preservation and Routine Passaging: Short-term maintenance of CmR laboratory strains on animal-free LB agar.
