LB Broth (Luria-Bertani) without Sodium Chloride is a nutrient medium containing tryptone and yeast extract, with sodium chloride omitted. It supports the cultivation of laboratory strains of E. coli and other non-fastidious bacteria under reduced ionic strength conditions.

The absence of NaCl provides low-salt conditions useful for salt-sensitive applications and downstream workflows. This broth serves as a base for routine propagation, plasmid maintenance, and selection when appropriate antibiotics are supplemented, and can be solidified with a gelling agent to prepare LB Agar without sodium chloride.

  • 1.0% Tryptone
  • 0.5% Yeast Extract
  • Cultivation of salt-sensitive strains: Growth of Escherichia coli and other bacteria when NaCl may inhibit growth or affect phenotype.

  • Plasmid propagation: Maintenance and amplification of plasmids in Escherichia coli under low-ionic-strength conditions.

  • Competent cell recovery: Post-transformation/electroporation recovery of Escherichia coli where reduced salt improves cell viability.

  • Bacteriophage work: Propagation and plaque assays (e.g., λ, M13) where low Na+ supports adsorption and infection dynamics.

  • Osmotic stress studies: Screening and characterization of osmosensitive mutants and salt-response phenotypes.

  • Protein expression: Expression in Escherichia coli for targets (e.g., membrane/ion-transport proteins) sensitive to Na+ levels.

  • General low-salt cultivation: Routine culture when reduced ionic strength is desired to minimize Na+-dependent effects.
