LB Agar Plates with Ampicillin-50 are solid media for the growth and selection of ampicillin-resistant E. coli strains. The inclusion of 50 µg/mL ampicillin sodium salt enables selection of plasmid-bearing transformants on a standard LB agar base.

  • 1.0% Tryptone
  • 0.5% Yeast Extract
  • 1.0% Sodium Chloride
  • 1.5% Agar
  • 50µg/mL Ampicillin Sodium Salt
  • Antibiotic Selection of Transformants: Selects for colonies carrying ampicillin-resistant plasmids after transformation of Escherichia coli.

  • Plasmid Maintenance on Solid Media: Maintains selection pressure to prevent plasmid loss during routine subculture of E. coli.

  • Colony Isolation and Purification: Streaks for single colonies to obtain clonal E. coli populations harboring Amp-resistant plasmids.

  • Colony Screening: Screens transformants by colony PCR or restriction analysis after picking Amp-resistant E. coli colonies.

  • Transformation Efficiency Testing: Quantifies CFUs to benchmark competent E. coli cell performance under Amp selection.

  • Cloning and Subcloning Workflows: Recovers recombinant plasmids post-ligation or Gibson assembly by selecting Amp-resistant E. coli clones.

  • Library Construction and Screening: Selects ampicillin-marked genomic, cDNA, or mutagenesis libraries in E. coli.

  • Conjugation and Mating Selections: Recovers transconjugants when the selectable marker confers ampicillin resistance in E. coli.

  • Expression Strain Recovery: Recovers and maintains expression plasmids with Amp resistance in E. coli prior to induction in liquid culture.

  • Quality Control of Plasmid Preparations: Verifies plasmid integrity and antibiotic marker function by re-plating on Amp with E. coli.
