LB Agar Plates with 6% Sucrose without Sodium Chloride are an LB-based solid medium for cultivation of E. coli and other enteric bacteria. The medium contains tryptone and yeast extract as nutrient sources, agar as the gelling agent, excludes sodium chloride, and includes sucrose as an additional carbon source suitable for sucrose-based counterselection workflows.

  • 1.0% Tryptone
  • 0.5% Yeast Extract
  • 1.5% Agar
  • 6.0% Sucrose

  • sacB Counter-Selection for Allelic Exchange: 6% sucrose enables lethal selection of sacB-bearing clones in Gram-negatives (e.g., Escherichia coli, Pseudomonas aeruginosa, Vibrio cholerae, Acinetobacter baumannii).

  • Agrobacterium tumefaciens Cultivation: LB without NaCl supports routine culture and selection workflows for plant transformation.

  • Spheroplast/Protoplast Recovery: High sucrose provides osmotic stabilization for plating and recovering fragile cells (e.g., transformed Escherichia coli).

  • Osmotic Stress Studies: High-osmolarity, NaCl-free medium for assessing osmoprotection, envelope stress, and compatibility solute responses.

  • Culturing NaCl-Sensitive Strains: Supports growth where sodium ions inhibit cells or interfere with antibiotics/assays.
