2 M Imidazole, pH 8.0 is a ready-to-use stock solution for preparing buffers and working solutions in protein purification and chromatographic workflows. Imidazole at this concentration is commonly used in immobilized metal affinity chromatography (IMAC) to modulate binding and to elute histidine-tagged proteins through competitive displacement.

The pH 8.0 specification provides compatibility with widely used aqueous buffer systems and is suited for dilution into Tris-, HEPES-, or phosphate-based buffers to achieve target imidazole concentrations for wash and elution steps.

  • 2M Imidazole

  • His-Tag Protein Purification (IMAC): Competitively elutes His-tagged proteins from Ni2+/Co2+ resins using 100–500 mM imidazole prepared from 2M Imidazole.

  • Binding/Wash Buffer Optimization: Reduces nonspecific binding during capture by adding 10–40 mM imidazole from 2M Imidazole to binding and wash buffers.

  • Affinity Pull-Downs of Protein Complexes: Uses controlled imidazole in wash/elution to isolate His-tagged complexes for proteomics.

  • On-Column Tag Cleavage Workflows: Maintains low imidazole during protease digestion to keep targets bound, followed by higher imidazole elution made from 2M Imidazole.

  • Buffering in Purification and Chromatography: Serves as a buffering component near neutral pH (≈7–8) in protein purification workflows, using sterile 2M Imidazole stock for rapid buffer preparation.
