1 M Imidazole, pH 8.0, is a ready-to-use aqueous solution commonly employed in immobilized metal affinity chromatography workflows for recombinant protein purification. At defined concentrations, imidazole is used to reduce nonspecific interactions during wash steps and to elute histidine-tagged proteins from nickel- or cobalt-charged resins, including proteins expressed in E. coli.

Imidazole at pH 8.0 is also used as a buffering component in biochemical assays and protein workflows where controlled imidazole levels are required. Typical use involves preparing wash and elution buffers by diluting the stock to the desired imidazole concentration for the specific target and resin system.

  • 1M Imidazole
  • Immobilized Metal Affinity Chromatography (IMAC): 1M Imidazole is diluted to create wash (10–40 mM) and elution (100–500 mM) buffers for His-tagged protein purification on Ni2+/Co2+ resins.

  • Wash Buffers for His-Tag Purification: Low concentrations from a 1M Imidazole stock reduce nonspecific binding of host proteins from Escherichia coli lysates during column washes.

  • Lysis and Binding Buffers: Added at 5–20 mM from a 1M Imidazole stock to suppress background binding while allowing His-tagged targets to capture on resin.

  • Step and Gradient Elution: Prepared from 1M Imidazole to generate precise step or linear gradients for optimized recovery of His-tagged proteins.

  • Denaturing IMAC: Used with urea or guanidine to purify insoluble His-tagged proteins; 1M Imidazole provides accurate high-concentration elutions under denaturing conditions.

  • Tag-Cleavage Workflows: After protease digestion (e.g., His-tagged TEV), 1M Imidazole-based buffers enable selective capture and elution of His-tagged species from flow-through mixtures.

  • Buffering in Biochemical Assays: Serves as a buffering component near neutral to mildly basic pH; 1M Imidazole stock is diluted to maintain pH ~7–8 in protein workflows.
