Isopropyl β-D-1-thiogalactopyranoside (IPTG) is a lactose analog used to induce transcription from lac operator–regulated promoters, including Plac, Ptac, Ptrc, and T7 systems controlled by LacI. IPTG binds the LacI repressor and derepresses target promoters, enabling controlled expression of recombinant genes in E. coli and other hosts carrying lac-derived regulatory elements.

This solution is suitable for addition to liquid cultures or agar media to initiate induction at defined cell densities or time points. Typical working concentrations range from 0.05 to 1 mM, with common use at 0.1–0.5 mM depending on promoter strength, strain background, and expression goals. Store at −20 °C protected from light; thaw as needed and minimize freeze–thaw cycles.

  • Expression Optimization Studies: Enables timing and dose–response optimization of lac-regulated expression using a 0.5M IPTG stock for precise final concentrations.

  • Metabolic Engineering and Pathway Induction: Controls lac-regulated operons for pathway flux studies and engineered metabolite production in E. coli.

  • Functional Studies of lac Operon Regulation: Probes lacI/lacO dynamics, inducer sensitivity, and promoter strength in E. coli.

  • Induction in Other lacI-Engineered Bacteria: Activates lac-controlled constructs in organisms engineered with lacI systems (e.g., Bacillus subtilis).
