Isopropyl β-D-1-thiogalactopyranoside (IPTG) is a non-metabolizable lactose analog used to induce transcription from lac operator–regulated promoters in bacteria. It binds the LacI repressor, facilitating expression from systems such as lac, tac, trc, and T7-lac promoters in E. coli and related hosts.

IPTG is commonly employed for controlled expression of recombinant proteins and in blue-white screening workflows in conjunction with chromogenic substrates. Its use enables regulated activation of operon-controlled genetic constructs without being consumed by cellular metabolism.

  • Isopropyl β-D-1-thiogalactopyranoside(IPTG)
  • Recombinant Protein Expression: Induces lac/tac/trc and T7-lac promoter systems for high-level protein production in Escherichia coli (e.g., BL21(DE3)).

  • Blue-White Colony Screening: Used with X-gal on selective plates to distinguish recombinant clones in E. coli.
