IPTG – Animal-Free is a non-metabolizable lactose analog used to induce transcription from lac operon–regulated promoters in bacteria such as E. coli. It binds the LacI repressor to enable controlled expression of genes under lac, tac, trc, and T7/lac control without being consumed by the cell.

This animal-free reagent is suitable for inducing recombinant protein expression and for blue–white screening in conjunction with X‑gal. It can be added to liquid cultures or agar media at defined concentrations (commonly 0.05–1 mM). IPTG is water‑soluble, compatible with filter sterilization, and is typically stored as an aqueous stock solution at −20 °C.

  • Isopropyl β-D-1-thiogalactopyranoside(IPTG)

  • Recombinant Protein Expression: Induces lac/T7-lac promoter-driven expression in Escherichia coli (E. coli) strains (e.g., DE3) for controlled protein production.

  • Blue-White Screening: Used with X-gal to induce lacZ for colony color selection in E. coli cloning workflows.

  • Reporter Gene Assays: Activates lac promoter–controlled reporters (e.g., lacZ) to quantify promoter activity in E. coli and other lac-based systems.

  • Genetic Circuit Control: Provides on-demand induction of genes under lac operators in synthetic biology constructs in E. coli.

  • CRISPR/Cas Module Induction: Drives expression of Cas or guide RNA components placed under lac or T7-lac promoters in E. coli.

  • Bacterial Two-Hybrid Systems: Induces interaction reporters controlled by lac-derived promoters for protein–protein interaction studies in E. coli.
