2X HBS (HEPES-Buffered Saline), pH 7.05, Endotoxin Tested, is a concentrated physiological buffer intended for preparation of 1X HBS by 1:1 dilution with water or compatible solutions. The buffering capacity is maintained at pH 7.05 to support pH stability in salt-based workflows and cell-based protocols.

Suitable for applications requiring HEPES-buffered saline at 2X strength or after dilution to 1X, including reagent preparation, washing steps, and the formation of calcium phosphate-DNA precipitates for transfection procedures.

  • 50mM HEPES
  • 280mM Sodium Chloride
  • 1.5mM Sodium Phosphate Dibasic
  • Calcium Phosphate Transfection: Optimized 2X buffer (50 mM HEPES, 280 mM NaCl, 1.5 mM Na2HPO4; pH 7.05) for forming CaPO4–DNA precipitates in mammalian cells.

  • Cell Culture Washing and Handling: Maintains physiological ionic strength and near-neutral pH during cell rinses, dilutions, and media exchanges.

  • Biosensor Running Buffer (SPR/BLI): Used as a base HEPES-saline buffer for protein–ligand interaction studies; often supplemented with surfactant/EDTA as needed.

  • Immunoassays (ELISA/Western Blot): Wash and dilution buffer where a HEPES-based saline is preferred over phosphate- or Tris-based systems.

  • Flow Cytometry/Immunostaining: Staining and wash buffer (commonly with BSA/azide) to preserve cell integrity and fluorescence signals.

  • Protein Purification Workflows: Equilibration/wash buffer for affinity and other chromatographic steps when phosphate-sensitive conditions are required.

  • Enzyme Assays: Provides stable pH near neutrality for activity measurements and inhibitor screening.

  • Viral Packaging Workflows: Buffer for transfection-driven production and subsequent washing steps in lentiviral/retroviral systems.
