1M HEPES, pH 7.0 is a zwitterionic buffering solution that maintains near-neutral pH in biological and biochemical applications. HEPES (4-(2-hydroxyethyl)-1-piperazineethanesulfonic acid) provides effective buffering in the pH range of approximately 6.8–8.2, with a pKa near 7.5 at 25 °C.

This solution is suitable for preparing working buffers by dilution for cell culture media, enzyme reactions, and handling of proteins and nucleic acids. It is used when a phosphate-free, Tris-free buffer is preferred and can be combined with salts or additives as required by the protocol.

  • Cell Culture Buffering: 1M HEPES is added to mammalian culture media to maintain physiological pH in low/no CO2 conditions.

  • Live-Cell Imaging: Stabilizes pH during fluorescence microscopy and time-lapse imaging of living cells.

  • Transfection/Viral Transduction: Used to prepare HEPES-buffered saline (HBS) and other transfection-compatible buffers.

  • Electrophysiology: Buffers extracellular and intracellular solutions for patch-clamp and ion channel studies.

  • Protein Purification/Storage: Maintains protein stability in chromatography buffers and storage solutions.

  • PCR/qPCR: Included in reaction or dilution buffers to maintain consistent pH during thermal cycling.

  • Flow Cytometry: Formulates HEPES-buffered staining and sorting buffers to support cell viability.

  • Gel Electrophoresis: Component of HEPES-based running or sample buffers for nucleic acid/protein separation.

  • Microbial Culture: Buffers defined media for Escherichia coli, Bacillus subtilis, and Pseudomonas aeruginosa to stabilize pH during growth.
