50% (v/v) Glycerol Solution is an aqueous glycerol preparation used as a cryoprotective additive and density modifier in laboratory workflows. It is commonly combined with microbial cultures to prepare freezer stocks for storage at low temperatures and to adjust sample density for handling and loading procedures.

In microbiology applications, this solution is used to prepare glycerol stocks of E. coli and other bacterial strains, supporting consistent archiving of working and reference cultures for subsequent revival and downstream analyses.

  • 50% (v/v) Glycerol
  • Glycerol Stocks for Bacteria: Preparation of long-term freezer stocks of Escherichia coli and other bacteria by diluting 50% (v/v) Glycerol to 10–25% final before freezing at −80°C.

  • Cryopreservation of Cells and Microorganisms: Use as a cryoprotectant (diluted from 50% (v/v) Glycerol) for yeasts, bacteria, and some mammalian cells to reduce ice damage during freezing.

  • Enzyme Storage: Stabilization of restriction enzymes, polymerases, and other proteins by storing in buffers containing up to 50% (v/v) Glycerol to remain unfrozen at −20°C.

  • Protein Stabilization: Addition to purification and storage buffers to prevent aggregation and denaturation of sensitive proteins.

  • PCR Enhancement: Inclusion of diluted 50% (v/v) Glycerol (typically 5–10% final) to improve amplification of difficult or GC‑rich templates.

  • Gel Electrophoresis Sample Loading: Added to samples as a density agent to ensure wells load cleanly and samples sink in agarose or PAGE gels.

  • Buffer Freeze Protection: Mixed into reagents to depress freezing point, enabling storage at −20°C without solidification.

  • Density Gradients/Cushions: Preparation of glycerol cushions or step gradients for ultracentrifugation and virus or ribosome pelleting.

  • Enzyme Assays: Used as an osmolyte/stabilizer in assay buffers to preserve enzymatic activity during measurements.
