20% (v/v) Glycerol Solution is an aqueous glycerol reagent commonly used as a cryoprotectant for long-term storage of microbial and cell cultures, as a protein and enzyme stabilizer, and as a solvent and density modifier in biochemical workflows.

Applications include preparation of glycerol stocks of E. coli and other bacteria, supplementation of storage buffers to reduce freeze–thaw damage, and adjustment of solution density for gradient-based separations.

  • 20% (v/v) Glycerol
  • Bacterial Glycerol Stocks: Cryoprotectant for long‑term storage of E. coli and other bacteria at −80°C; mix culture with 20% (v/v) Glycerol to achieve a typical final 10–20% (v/v).

  • Electrocompetent Cell Preparation: Preparation of electrocompetent E. coli using ice-cold glycerol washes (e.g., final ~10%); dilute 20% (v/v) Glycerol 1:1 for working solutions.

  • Media Supplementation (Carbon Source): Sterile addition to media to reach desired final 0.2–1% (v/v) glycerol for growth of E. coli, Pseudomonas, and other microbes.

  • Protein and Enzyme Stabilization: Added to storage and assay buffers (final 5–20%) to reduce aggregation and freeze–thaw damage.

  • Gel Loading Density Adjustment: Increases density of DNA/RNA/protein samples for electrophoresis (final 5–15%); 20% (v/v) Glycerol used to titrate density.

  • Glycerol Gradient Centrifugation: Component for preparing 10–30% glycerol gradients to fractionate protein and nucleic acid complexes.

  • PCR and Restriction/Ligation Additive: Low-percentage glycerol (2–10%) improves enzyme stability/specificity; 20% (v/v) Glycerol used for accurate dilution.

  • Yeast and Fungal Cryopreservation: Cryoprotectant for long-term storage of strains such as Saccharomyces cerevisiae at −80°C.

  • Bacteriophage Preservation: Cryoprotectant for phage lysates propagated in hosts like E. coli.
