500 mM EDTA, pH 7.5, is an aqueous chelating solution used to sequester divalent metal ions such as Mg2+ and Ca2+ in molecular biology and biochemistry workflows.

Applications include incorporation into buffers (e.g., TE) for nucleic acid work, inhibition of metal-dependent nucleases, cell lysis and extraction procedures, and washing steps in microbiology involving E. coli.

  • 500mM Ethylenediaminetetraacetic acid (EDTA)
  • Nucleic Acid Preservation (TE Buffer): Chelates Mg2+ to inhibit DNases/RNases; used to store and stabilize DNA/RNA.

  • Agarose Gel Electrophoresis Buffers (TAE/TBE): Provides EDTA to protect nucleic acids from nuclease activity during electrophoresis.

  • Quenching Mg2+-Dependent Reactions: Stops polymerase, ligase, and restriction enzyme reactions by sequestering divalent cations.

  • Protein Purification/Storage: Inhibits metalloproteases and chelates trace metals to prevent protein degradation and oxidation.

  • IMAC Resin Stripping/Regeneration: Removes Ni2+/Co2+ from charged affinity resins during cleanup or regeneration.

  • Cell Dissociation in Tissue Culture: Used in Ca2+/Mg2+-free buffers (e.g., PBS-EDTA) to gently detach adherent cells.

  • Competent Cell Preparation: Chelates divalent cations to permeabilize outer membranes in Escherichia coli workflows.

  • Biofilm and Membrane Studies: Disrupts Ca2+-dependent biofilms and increases outer membrane permeability in Pseudomonas aeruginosa.

  • DNA/RNA Sample Transport: Added to collection/stabilization buffers to preserve integrity of nucleic acids.

  • Tissue Decalcification (Histology): Slowly chelates calcium from mineralized tissues for morphology-preserving decalcification.
