500 mM EDTA, pH 8.0, is an aqueous chelating solution for binding divalent metal ions such as Mg2+, Ca2+, Mn2+, and Zn2+. The pH 8.0 specification provides the deprotonated form used to sequester metal cofactors in solution.

Applications include limiting metal-dependent nuclease activity during DNA and RNA handling, preparing and adjusting buffers (e.g., TE), conditioning reagents and water by removing trace metals, and controlling divalent cation availability in biochemical and microbiological workflows involving organisms such as E. coli.

  • 500mM Ethylenediaminetetraacetic acid (EDTA)
  • Nuclease Inhibition: Chelates Mg2+ and Ca2+ to inhibit DNases/RNases during DNA/RNA extraction and handling.

  • TE Buffer Component: Combined with Tris to formulate TE for long-term DNA/RNA storage and stability.

  • Plasmid Preparation: Included in resuspension/lysis buffers to protect plasmid DNA from nucleases.

  • PCR/qPCR Sample Preservation: Added to samples to chelate Mg2+, suppress nuclease activity, and quench reactions post-amplification.

  • Protease Inhibition: Inhibits metalloproteases in cell lysates by sequestering divalent metal cofactors.

  • Protein Buffering/Chelation: Prevents metal-catalyzed oxidation and aggregation in protein storage and assay buffers.

  • Competent Cell Preparation: Facilitates outer membrane permeabilization of Escherichia coli by chelating divalent cations.

  • Divalent Cation Removal: Eliminates trace Mg2+/Ca2+ from solutions, glassware, and reagents to control metal-dependent reactions.

  • Decalcification: Chelates calcium to decalcify tissues and dissolve biomineral deposits.

  • IMAC Resin Stripping: Removes Ni2+/Co2+ from immobilized metal affinity chromatography resins during regeneration.
