EMB Agar Plates are selective and differential microbiological media used for isolation and differentiation of Gram-negative enteric bacteria based on lactose fermentation. The combination of eosin Y and methylene blue provides pH-responsive coloration and partial inhibition of Gram-positive organisms.

Lactose-fermenting colonies typically appear dark purple to black, with a metallic green sheen often observed for some strains of Escherichia coli. Non–lactose-fermenting colonies (e.g., Salmonella, Shigella) are usually colorless to light pink.

  • 1.00% Peptone
  • 0.20% Dipotassium Phosphate
  • 0.04% Eosin Y Disodium Salt
  • 1.50% Agar
  • 65µg/mL Methylene Blue Hydrate
  • 2.00% Lactose
  • Isolation and Differentiation of Enteric Gram-Negatives: Selective/differential plating to distinguish lactose fermenters (e.g., metallic green sheen of Escherichia coli) from non-fermenters using 2.00% Lactose and the Eosin Y/Methylene Blue dye system.

  • Coliform and Fecal Coliform Detection (Water/Wastewater): Confirmation plating for total and fecal coliforms following enrichment or membrane filtration to assess potable and environmental water quality.

  • Food and Beverage Testing: Enumeration and confirmation of coliforms as hygiene indicators in dairy, meat, produce, and beverages to verify sanitation and process control.

  • Clinical Stool Culture Screening: Differentiation of lactose-negative pathogens such as Salmonella and Shigella from normal flora, while identifying lactose-positive coliforms like Escherichia coli.

  • Urinary Tract Infection Workups: Isolation and preliminary differentiation of Gram-negative rods, including Escherichia coli, Klebsiella pneumoniae, and Proteus, based on lactose fermentation.

  • Environmental and Cleanroom Monitoring: Recovery of Gram-negative contaminants from surfaces, air, and process samples in pharmaceutical, biotech, and healthcare settings.

  • Teaching and Training: Demonstration of selective/differential media principles and classic colony morphologies (e.g., Escherichia coli green sheen) in academic labs.

  • Colony Purification Prior to ID: Isolation of single colonies for downstream biochemical identification, MALDI-TOF, or molecular assays.
