Denaturation Solution for Southern Hybridization is an alkaline high-salt reagent used to convert double-stranded DNA to single-stranded form on nylon or nitrocellulose membranes prior to probe hybridization. It is applied after DNA transfer from agarose gels and before neutralization and hybridization steps in standard Southern blot workflows.

This solution is suitable for genomic DNA analysis using labeled nucleic acid probes and is compatible with capillary or vacuum transfer methods. Store at room temperature tightly closed and use with appropriate neutralization and wash buffers as required by the assay protocol.

  • 1.5M Sodium Chloride
  • 0.5M Sodium Hydroxide
  • Southern Blot Gel Denaturation: Alkaline denaturation of double-stranded DNA in agarose gels prior to capillary transfer.

  • Membrane-Bound DNA Denaturation: Denatures DNA immobilized on nylon or nitrocellulose membranes before probe hybridization.

  • Alkaline Capillary Transfer: Used as the denaturation/transfer solution in alkaline Southern blot protocols to enhance DNA transfer efficiency.

  • Dot/Slot Blot Preparation: Denatures plasmid or genomic DNA applied to membranes to enable single-stranded target formation for probe binding.

  • Colony and Plaque Lifts: Denatures DNA on lifts from E. coli colonies or bacteriophage plaques to prepare targets for hybridization.

  • RNA Background Reduction on DNA Blots: Alkaline conditions hydrolyze RNA contaminants, lowering nonspecific signal.

  • Probe Accessibility Enhancement: Converts dsDNA to ssDNA on gels/membranes to improve hybridization kinetics and signal intensity.

  • DNA Immobilization Verification: Applied in test blots to confirm efficient fixation and retention of denatured DNA on membranes.
