CM Raffinose Broth minus Uracil is a defined liquid medium for cultivation and selection of URA3-positive strains of Saccharomyces cerevisiae under uracil dropout conditions. Omission of uracil enables maintenance of URA3-marked plasmids or genomic integrations in ura3 backgrounds.

Raffinose is provided as the primary carbon source and is compatible with galactose-inducible expression systems; ammonium sulfate supplies inorganic nitrogen. The formulation includes yeast nitrogen base and a complete supplement mixture lacking uracil; add required amino acids and bases according to the auxotrophic genotype, excluding uracil.

  • 0.5% Ammonium Sulfate
  • URA3 Selection in Yeast Transformation: Selects and enriches Saccharomyces cerevisiae URA3+ transformants in liquid culture using uracil dropout with 2.0% Raffinose and 0.5% Ammonium Sulfate.

  • Plasmid Maintenance and Expansion: Maintains URA3-marked plasmids in Saccharomyces cerevisiae during overnight cultures in raffinose-based (-Ura) broth for high-yield biomass.

  • GAL Promoter Workflows: Pre-induction growth in 2.0% Raffinose to avoid glucose repression prior to galactose induction of GAL-driven expression.

  • Yeast Two-Hybrid and Library Screens: Supports liquid -Ura selection for bait/prey plasmid maintenance and assay setup under defined nitrogen (0.5% Ammonium Sulfate).

  • Plasmid Shuffling and Counterselection Prep: Grows URA3-dependent strains in -Ura broth before downstream 5-FOA counterselection steps.

  • Carbon Source and Growth Kinetics Studies: Evaluates respiratory growth and sugar utilization on 2.0% Raffinose with controlled nitrogen via 0.5% Ammonium Sulfate.
