CM Glucose Broth minus Leucine and Uracil is a defined yeast culture medium formulated without leucine and uracil to enable selection of auxotrophic markers (LEU2 and URA3) in Saccharomyces cerevisiae. Glucose serves as the primary carbon source, and ammonium sulfate provides inorganic nitrogen for biosynthesis.

This medium is suitable for propagation, plasmid maintenance, and dropout selection workflows in yeast genetics when combined with appropriate supplements or selectable markers targeting leucine and uracil pathways.

  • 2.0% Cerelose Anhydrous Dextrose (Glucose)
  • 0.5% Ammonium Sulfate
  • Yeast Auxotrophic Selection: Selects and maintains Saccharomyces cerevisiae transformants carrying LEU2 and URA3 markers in a defined medium with 2.0% glucose and 0.5% ammonium sulfate.

  • Dual-Plasmid Maintenance: Co-maintenance of LEU2/URA3 plasmids for cloning, expression, and stability studies in yeast.

  • Yeast Two-Hybrid and Interaction Assays: Supports assays requiring LEU2 and URA3 selection for bait/prey or reporter constructs.

  • CRISPR/Cas9 Genome Editing: Selection of edited yeast using LEU2/URA3 markers and workflows preceding URA3 counterselection (e.g., 5-FOA).

  • Transformation and Recovery: Outgrowth and enrichment of yeast transformants under -Leu -Ura selection.

  • Plasmid Shuffling and Retention/Loss Assays: Maintenance or eviction of LEU2/URA3-marked plasmids, including pre-5-FOA culture steps.

  • Growth and Phenotypic Assays in Defined Conditions: Controlled studies of growth, starvation responses, and gene expression with 2.0% glucose as carbon and 0.5% ammonium sulfate as nitrogen source.
