CM Glucose Broth minus Uracil is a defined minimal medium that provides glucose as the carbon source and omits uracil.

It is used for cultivation and selection of uracil-prototrophic yeast (e.g., Saccharomyces cerevisiae) and for maintenance of strains or plasmids bearing URA3 in uracil-dropout conditions.

  • 2.0% Cerelose Anhydrous Dextrose (Glucose)
  • 0.5% Ammonium Sulfate
  • Auxotrophic Selection (-Ura): Selects and maintains URA3+ transformants of Saccharomyces cerevisiae in liquid culture by omitting uracil, using 2.0% glucose and 0.5% ammonium sulfate as defined carbon and nitrogen sources.

  • Plasmid Maintenance (URA3 Marker): Supports stable propagation of URA3-marked plasmids in yeast during cloning, expression, and strain construction.

  • Yeast Two-Hybrid Screens: Enables selection of reporter strains and plasmids requiring uracil dropout in liquid assays.

  • CRISPR/Cas9 Editing Workflows: Facilitates selection of edited yeast cells using URA3-based selectable markers in defined broth.

  • Library Selections and Enrichment: Performs pooled selections for barcoded or display libraries relying on URA3 auxotrophy.

  • Phenotypic Assays in Defined Media: Assesses growth, metabolite production, and stress responses under controlled 2% glucose/0.5% ammonium sulfate conditions without uracil.

  • Growth Curves and Kinetic Studies: Measures yeast growth rates and drug sensitivity in uracil-dropout broth.

  • Replica Plating and Dropout Validation: Confirms genotype (URA3 dependence) and plasmid retention following selection.

  • Protein Expression in Yeast: Cultures URA3+ expression strains for recombinant protein production in defined liquid media.

  • Genetic Crosses and Segregant Analysis: Selects desired meiotic progeny carrying URA3-linked traits in liquid selection steps.

  • pH: 5.70 ± 0.10
