CM Glucose Broth Minus Tryptophan is a carbohydrate-based liquid medium formulated without tryptophan for cultivation and characterization of microorganisms under tryptophan-limited conditions. Glucose is supplied as Cerelose anhydrous dextrose to provide a defined carbon source, and ammonium sulfate supplies inorganic nitrogen.

This formulation is suitable for evaluating tryptophan auxotrophy, monitoring glucose-driven growth and metabolite production, and maintaining cultures where tryptophan omission is required by the experimental design. It can be used with organisms such as E. coli that are prototrophic for tryptophan or with engineered strains in studies of amino acid biosynthesis, transport, and regulation.

  • 2.0% Cerelose Anhydrous Dextrose (Glucose)
  • 0.5% Ammonium Sulfate
  • Yeast Auxotrophic Selection (-Trp): Selects for TRP1-marked plasmids/strains in Saccharomyces cerevisiae using a 2.0% Cerelose Anhydrous Dextrose (Glucose), 0.5% Ammonium Sulfate base.

  • Yeast Two-Hybrid (Y2H) Bait Selection: Primary selection of bait constructs on -Trp medium prior to interaction screening.

  • Maintenance of TRP1-Tagged Transformants: Stable propagation of yeast carrying TRP1 plasmids without additional supplements.

  • Synthetic Genetic Array (SGA) and Interaction Screens: Marker-based selection steps requiring -Trp dropout to track genetic constructs.

  • Replica Plating and Spot Assays: Genotype confirmation and fitness assays under tryptophan dropout.

  • Metabolic and Starvation Studies: Assess growth, tryptophan biosynthesis/uptake, and stress responses in -Trp, glucose-repressed conditions.

  • GAL Promoter Repression Pre-Cultures: 2.0% glucose supports robust growth while repressing GAL1/10-driven expression prior to induction.
