Chocolate Agar Plates are enriched, nonselective culture media formulated from GC agar base with hemoglobin and an enrichment supplement. Heating lysed red blood cells provides hemin X factor and releases nicotinamide adenine dinucleotide V factor to support the cultivation of fastidious organisms.

This medium is routinely used for the isolation and cultivation of clinical isolates including Haemophilus influenzae, Haemophilus parainfluenzae, Neisseria gonorrhoeae, and Neisseria meningitidis, as well as other fastidious respiratory and oropharyngeal bacteria in diagnostic workflows.

  • 3.6% GC Agar Base
  • 1.0% IsoVitalex Enrichment
  • 1.0% Hemoglobin
  • 1.5% Agar
  • Primary Isolation of Fastidious Respiratory Pathogens: Nonselective enriched recovery of Haemophilus influenzae and Haemophilus parainfluenzae from nasopharyngeal, sputum, and BAL specimens.

  • Isolation of Pathogenic Neisseria: Cultivation of Neisseria gonorrhoeae and Neisseria meningitidis from urogenital, pharyngeal, and cerebrospinal fluid specimens.

  • Recovery of X and V Factor–Dependent Organisms: Supports growth of Haemophilus spp. requiring hemin (X) and NAD (V) supplied by hemoglobin and enrichment.

  • Cultivation Under CO2-Enriched Conditions: Enhanced recovery of fastidious organisms (e.g., Neisseria, Haemophilus) incubated in 5–10% CO2.

  • Processing Mixed Clinical Specimens: Enriched medium to increase yield of target fastidious bacteria from polymicrobial samples.

  • Preliminary Identification Workflows: Supports colony morphology assessment and rapid oxidase testing for Neisseria and catalase/indole testing for Haemophilus.

  • Quality Control and Method Validation: Routine growth control of fastidious reference strains such as H. influenzae and N. gonorrhoeae.
