CM Glucose Agar Plates minus Uracil are a defined agar medium formulated with glucose as the primary carbon source and ammonium sulfate as the inorganic nitrogen source. The medium is prepared as ready-to-use plates for cultivation of yeast strains on a minimal, chemically defined base.

Omission of uracil enables selection and maintenance of uracil-auxotrophic strains complemented by URA-marked plasmids, and supports screening of genetic constructs relying on URA3-based selection. The medium is suitable for work with Saccharomyces cerevisiae and related yeasts requiring defined conditions for auxotrophic selection.

  • 2.0% Cerelose Anhydrous Dextrose (Glucose)
  • 0.5% Ammonium Sulfate
  • URA3-Based Selection in Yeast Genetics: Positive selection of URA3+ transformants and integrants on defined CM medium with 2.0% Cerelose (glucose) and 0.5% ammonium sulfate.

  • Plasmid Maintenance: Stable maintenance and recovery of URA3-marked plasmids in auxotrophic yeast strains.

  • Yeast Two-Hybrid/Reporter Assays: Selection of reporter constructs utilizing URA3, supporting defined carbon and nitrogen sources for consistent readouts.

  • CRISPR/Cas9 Genome Editing: Screening edits using URA3 as a selectable marker on CM glucose agar lacking uracil.

  • Genetic Crosses and Tetrad Analysis: Identification of URA3 prototrophs during segregation analysis on defined medium.

  • Library Screens: Enrichment of genomic or cDNA libraries carried on URA3-marked vectors.

  • Phenotypic Characterization: Assessing growth and metabolic phenotypes under defined 2.0% Cerelose (glucose) and 0.5% ammonium sulfate conditions without uracil.

  • Counter-Selection Workflows: Used as the positive-selection counterpart in 5-FOA-based URA3 counter-selection strategies.
