CM Glucose Agar Plates minus Tryptophan and Uracil are defined agar media in which glucose is the sole carbon source and ammonium is the inorganic nitrogen source. The omission of tryptophan and uracil enables selection and maintenance of yeast strains or plasmids carrying TRP1 and URA3 markers under auxotrophic conditions.

Formulated for routine cloning workflows and genetic analyses in Saccharomyces cerevisiae, these plates support growth only when the required biosynthetic functions are complemented. Compatible with standard yeast dropout supplements for targeted amino acid and nucleotide base control.

  • 2.0% Cerelose Anhydrous Dextrose (Glucose)
  • 0.5% Ammonium Sulfate
  • Yeast Auxotrophic Selection (-Trp -Ura): Selects for TRP1- and URA3-complemented S. cerevisiae strains using 2.0% Cerelose Anhydrous Dextrose (Glucose) and 0.5% Ammonium Sulfate as defined carbon and nitrogen sources.

  • Plasmid Maintenance: Co-selects and maintains TRP1- and URA3-marked plasmids in yeast during cloning and propagation.

  • Yeast Two-/One-Hybrid Workflows: Supports co-transformation and selection when bait/prey or reporter vectors use TRP1 and URA3 markers.

  • CRISPR-Cas9 Genome Editing: Enables selection of edits or donor integration using URA3/TRP1 selectable markers.

  • Transformation Screening: Facilitates colony recovery and selection of correctly transformed yeast on dropout media.

  • Plasmid Shuffling and Counter-Selection Setups: Used for initial URA3 selection prior to 5-FOA counter-selection strategies.

  • Replica Plating and Genotype Verification: Distinguishes trp1Δ and ura3Δ backgrounds and confirms plasmid-dependent growth.

  • Phenotypic and Drug Sensitivity Assays: Maintains selection pressure while assessing growth phenotypes on defined glucose media.

  • Library and Pathway Screening: Screens combinatorial plasmid libraries requiring simultaneous TRP1 and URA3 selection.

  • Glucose-Repression Studies: Uses 2.0% Cerelose (glucose) with 0.5% Ammonium Sulfate to analyze carbon source–dependent regulation under defined conditions.
