CM Glucose Agar Plates minus Tryptophan and Uracil are defined agar media designed for selection and maintenance of auxotrophic yeast. Glucose serves as the primary carbon source, and ammonium sulfate provides inorganic nitrogen, while omission of tryptophan and uracil enables selection for TRP1 and URA3 prototrophy in Saccharomyces cerevisiae.

This medium supports cloning workflows, plasmid maintenance, and genotype verification in yeast strains carrying complementing markers, and is suitable for routine streaking, colony isolation, and replica plating.

  • 2.0% Cerelose Anhydrous Dextrose (Glucose)
  • 0.5% Ammonium Sulfate
  • Auxotrophic Selection (TRP1/URA3): Selects and maintains Saccharomyces cerevisiae strains or plasmids carrying TRP1 and URA3 markers by omitting tryptophan and uracil.

  • Yeast Transformation & Plasmid Maintenance: Post-transformation selection and stable maintenance of dual-marked vectors (TRP1, URA3) in S. cerevisiae.

  • CRISPR/Cas9 Editing Workflows: Marker-based selection of genome edits using TRP1 or URA3 selectable cassettes.

  • Library Screening & Clone Picking: High-throughput screening of yeast genomic or cDNA libraries under defined dropout conditions.

  • Yeast Two-Hybrid/Reporter Pipelines: Support assays requiring TRP1/URA3 selection for bait/prey or reporter plasmids.

  • Replica Plating & Segregation Analysis: Track genetic markers and plasmid retention across colonies on defined dropout medium.

  • Strain Construction & Backcrossing: Build and verify strains with TRP1/URA3 auxotrophies or plasmid complements.

  • Metabolic and Nutrient Utilization Studies: Defined 2.0% Cerelose Anhydrous Dextrose (Glucose) and 0.5% Ammonium Sulfate enable controlled carbon/nitrogen studies in yeast.

  • Phenotypic Assays under Defined Conditions: Evaluate growth, stress responses, and compound sensitivity with precise dropout and standardized glucose/ammonium levels.
