CCMB80 Buffer is a reagent used in the preparation of chemically competent E. coli cells for heat-shock transformation workflows. It contains divalent cation salts and potassium acetate in a glycerol-based solution and is used as a wash and resuspension buffer during cell competency protocols.

The combination of calcium, manganese, and magnesium salts provides the ionic conditions referenced in classical chemical transformation methods for E. coli, while glycerol serves as an osmoprotectant during chilling and handling. CCMB80 Buffer is suitable for use in routine cloning procedures that require preparation of chemically competent E. coli.

  • 80mM Calcium Chloride
  • 20mM Manganese Chloride

  • 10% Glycerol
  • Preparation of Chemically Competent E. coli: Used to generate high-efficiency competent cells; Ca2+, Mn2+, and Mg2+ promote DNA uptake while potassium acetate and glycerol stabilize cells.

  • Heat-Shock Transformation: Supports efficient uptake of plasmids, ligation products, Gibson/Golden Gate assemblies into E. coli.

  • High-Throughput Library Construction: Enables transformation for large plasmid and sgRNA libraries requiring high competence in E. coli.

  • Routine Cloning and Strain Engineering: Facilitates cloning, site-directed mutagenesis, and CRISPR plasmid introduction into E. coli.

  • Competent Cell Storage: 10% glycerol provides cryoprotection for short-term storage of competent E. coli at −80°C.

  • Processing Buffer During Competent Cell Preparation: Used for washing and resuspension steps to maintain osmotic balance and membrane stability.

  • Transformation of Recalcitrant E. coli Strains: Improves transformation efficiency in strains that are difficult to make competent.
