CM Glucose Plates minus Tryptophan are a defined agar medium that supports growth and selection of tryptophan-prototrophic yeast strains. Glucose serves as the primary carbon source and ammonium sulfate provides inorganic nitrogen, while the omission of tryptophan enables selection for TRP1-based auxotrophic complementation.

Designed for maintenance and selection of plasmid-bearing transformants in Saccharomyces cerevisiae using TRP1 markers.

  • 2.0% Glucose
  • 0.5% Ammonium Sulfate
  • Minus: Tryptophan
  • Yeast Auxotrophic Selection (-Trp): Selects TRP1-prototrophic transformants on defined 2.0% Glucose, 0.5% Ammonium Sulfate plates lacking tryptophan.

  • Maintenance of TRP1-Marked Plasmids: Ensures stable retention of TRP1 vectors in yeast by growth on -Trp plates.

  • Yeast Two-Hybrid Bait Selection: Selects bait constructs carrying TRP1 prior to interaction screening (e.g., -Leu/-Trp for bait-prey).

  • Yeast Transformation and Cloning Workflows: Enables colony recovery and screening of transformants with TRP1 markers on -Trp selective medium.

  • CRISPR/Cas9 Editing with TRP1 Selection: Recovers edited clones using TRP1 cassettes or donor constructs on tryptophan dropout plates.
