Agar, Molecular Biology Grade is a plant-derived polysaccharide gelling agent used to solidify liquid media for the cultivation, maintenance, and selection of microorganisms, including E. coli and yeast, and for routine molecular biology workflows such as cloning and transformation. It enables preparation of plates, slants, and overlays for a range of culture formats.

This agar dissolves upon heating and resolidifies on cooling to form stable gels across typical incubation temperatures. It can be sterilized by autoclaving and is compatible with common media and supplements added at appropriate temperatures, including antibiotics, sugars, salts, and chromogenic substrates.

Applications include preparation of LB, 2×YT, and minimal media plates; selection of transformants with antibiotic-containing media; blue–white screening with X‑gal/IPTG; bacteriophage plaque assays using soft agar overlays; motility assays using reduced gel strength; and long-term maintenance of bacterial and yeast stocks, including E. coli and Saccharomyces cerevisiae.

  • Bacteriophage Plaque Assays: Used as soft/top agar overlays for phage propagation and titration on bacterial lawns (e.g., Escherichia coli).

  • Motility and Chemotaxis Assays: Low-percentage agar enables assessment of bacterial motility (e.g., Escherichia coli, Salmonella enterica).

  • Replica Plating and Colony Lifts: Transfers colonies between selective media to screen mutants or gene knockouts in Escherichia coli and other bacteria.

  • Microbial Enumeration (CFU Counts): Pour or spread plates for quantitative recovery of bacteria from water, food, and environmental samples.

  • Antimicrobial Susceptibility Testing: Agar-based diffusion and gradient assays to evaluate bacterial sensitivity to antibiotics.

  • Yeast and Fungal Cultivation: Gelling agent for media used to grow yeasts and filamentous fungi.

  • Plant and Model Organism Media: Gelling agent for plant tissue culture (e.g., MS media) and NGM plates for C. elegans.
