5X Reaction Buffer is a concentrated buffer for ATP-dependent enzymatic workflows. It provides ATP and magnesium as cofactors, Tris-HCl for pH control, and sodium chloride to maintain ionic strength. TCEP maintains a reducing environment, and Tween-20 limits protein adsorption to surfaces. When diluted to working strength, it supports reactions catalyzed by nucleotidyl transferases, kinases, ligases, helicases, and other ATP-utilizing enzymes.

  • 10mM ATP
  • 800mM Sodium Chloride
  • 25mM Magnesium Chloride
  • 12.5mM TCEP
  • 1% Tween-20
  • 150mM Tris-HCl
  • Protein Kinase Assays: Supports phosphorylation reactions with 10 mM ATP and 25 mM MgCl2 for optimal kinase activity in a 150 mM Tris-HCl pH environment.

  • ATPase Activity Measurements: Enables motor/chaperone ATPase assays with ATP and Mg2+, while 1% Tween-20 minimizes surface adsorption.

  • DNA/RNA Helicase Unwinding Assays: Provides ATP and Mg2+ for helicase function, with controlled ionic strength from NaCl for substrate stability.

  • ATP-Dependent Ligase Reactions: Supplies ATP and Mg2+ required for DNA/RNA ligation, with TCEP maintaining a reducing environment.

  • Proteasome and AAA+ Enzyme Assays: Supports ATP-dependent unfolding/degradation activities with appropriate Mg2+ and salt conditions.

  • Chromatin Remodeling Reactions: Facilitates ATP-driven remodeler activity with defined Tris-HCl buffering and NaCl for nucleoprotein complex stability.

  • Nucleic Acid Translocase/RecA/RadA Assays: Provides ATP/Mg2+ for strand exchange/translocation with TCEP to protect protein thiols.

  • Type III Restriction–Modification Reactions: Supplies ATP for ATP-dependent restriction enzymes, with Mg2+ as a required cofactor.

  • Enzyme Stabilization and Dilution: TCEP and 1% Tween-20 reduce oxidation and nonspecific adsorption during handling and storage.

  • General ATP-Dependent Biochemical Reactions: 150 mM Tris-HCl for pH control and 160 mM NaCl at 1X (from 800 mM at 5X) provide balanced conditions across diverse ATP-driven enzymes.
