qPCR Proteinase K Buffer is a high-salt, anionic surfactant–containing buffer formulated to support robust Proteinase K–mediated lysis and protein denaturation across a range of biological sample types. The composition facilitates disruption of cellular and viral membranes, solubilization of proteins, and reduction of nuclease activity, aiding preservation of nucleic acids during extraction.

The buffer is suitable for workflows that include Proteinase K digestion followed by downstream quantitative PCR analysis. It can be used with tissues, swabs, and other clinical or environmental specimens, and is compatible with standard Proteinase K concentrations and thermal inactivation or dilution steps commonly used prior to amplification.

  • 1.11M Sodium Chloride
  • 1.11% Sarkosyl
  • qPCR Sample Preparation: Facilitates Proteinase K–mediated lysis and nucleic acid release from swabs, saliva, and tissues; 1.11M Sodium Chloride and 1.11% Sarkosyl enhance cell disruption and nuclease inactivation for downstream qPCR.

  • DNA/RNA Extraction: Detergent/salt-assisted protein denaturation and membrane solubilization to improve yield and purity of nucleic acids prior to qPCR quantification.

  • Microbial Lysis for qPCR: Rapid lysis of bacteria such as Escherichia coli and Staphylococcus aureus to enable sensitive detection and quantification by qPCR.

  • Viral Inactivation and Stabilization: Sarkosyl-based disruption aids inactivation of enveloped viruses and stabilizes released nucleic acids for reliable qPCR readouts.

  • Nuclease Inactivation: High salt (1.11M NaCl) and detergent (1.11% Sarkosyl) conditions help denature RNases/DNases, protecting target nucleic acids.

  • Tissue Digestion: Supports Proteinase K digestion of protein-rich or viscous samples (e.g., sputum, tissue homogenates) to reduce inhibitors and improve qPCR performance.

  • Inhibitor Reduction: Protein and lipid solubilization decreases PCR inhibitors, improving amplification efficiency and Ct consistency.

  • Environmental and Food Testing: Lysis of microbial cells from water, surfaces, and food matrices to prepare samples for contamination assessment by qPCR.
