Modified Columbia Agar with 5% Sheep’s Blood is an enriched, blood-supplemented agar medium used for the cultivation and isolation of fastidious bacteria and for observation of hemolytic reactions. The addition of defibrinated sheep’s blood enables visualization of alpha, beta, and gamma hemolysis and supports assessment of colony morphology directly on the primary isolation plate.

This medium is routinely applied for recovery and presumptive differentiation of organisms such as Streptococcus spp. and Staphylococcus aureus from clinical and other specimen types under aerobic or CO₂-enriched conditions.

  • 1.20% Tryptone
  • 0.50% Peptone
  • 0.35% Yeast Extract
  • 0.30% Beef Extract
  • 0.10% Corn Starch
  • 0.50% Sodium Chloride
  • 1.50% Agar
  • 5.00% Sheep's Blood
  • Isolation of Hemolytic Bacteria: Differentiates alpha, beta, and gamma hemolysis for organisms such as Streptococcus pyogenes, Streptococcus pneumoniae, and Staphylococcus aureus.

  • Cultivation of Fastidious Organisms: Supports growth of nutritionally demanding bacteria including Streptococcus spp., Enterococcus spp., and Listeria monocytogenes.

  • Clinical Specimen Culture: Primary plating medium for throat, wound, and respiratory specimens to recover diverse pathogens.

  • Hemolysin Detection: Visualizes hemolytic toxin production for characterization of Streptococcus and Listeria species.

  • Antimicrobial Susceptibility Screening: Used for disk diffusion or gradient strip testing of fastidious bacteria when blood-enriched media are required.

  • Mixed Flora Assessment: Enables observation of colony morphology and hemolysis in polymicrobial samples.

  • Anaerobe Recovery (with Anaerobic Incubation): Supports growth of obligate anaerobes such as Clostridium spp. and Bacteroides spp. under appropriate conditions.

  • Quality Control and Performance Testing: Routine QC with reference strains (e.g., Streptococcus pyogenes for beta hemolysis, Streptococcus pneumoniae for alpha hemolysis).
