Assay Diluent is a buffered saline containing a protein carrier and a nonionic surfactant for preparation and dilution of samples, standards, and conjugates in immunoassays, including ELISA and bead-based formats.

The buffer maintains near-physiological ionic strength and pH, the surfactant aids wetting and dispersion, and the protein component serves as a carrier to stabilize proteins and limit surface adsorption.

  • 10mM Tris-HCl
  • 0.90% Sodium Chloride
  • 0.05% (v/v) Tween-20
  • 1.00% Bovine Serum Albumin (BSA)
  • ELISA Sample Dilution: Dilutes serum, plasma, or cell culture supernatants while minimizing matrix effects and nonspecific binding.

  • Antibody Diluent (ELISA/EIA): Stabilizes capture and detection antibodies and reduces background during incubations.

  • Western Blot Antibody Dilution: Serves as a primary/secondary antibody diluent to lower nonspecific interactions and improve signal-to-noise.

  • Plate Blocking/Incubation Buffer: Supplements or replaces separate blocking steps to decrease hydrophobic binding on polystyrene plates.

  • Lateral Flow and Rapid Immunoassays: Used as a sample/conjugate diluent to enhance specificity and maintain protein stability.

  • Bead-Based Multiplex Assays (e.g., Luminex): Provides consistent protein-friendly conditions for bead incubations and standard curves.

  • Immunohistochemistry/Immunofluorescence: Dilutes primary/secondary antibodies to reduce off-target binding in tissue and cell staining.

  • Protein Standards and Calibrators: Maintains analyte stability and linearity for standard curve preparation.

  • General Protein Stabilization: Temporarily stabilizes antigens, enzymes, and conjugates during assay setup and handling.

  • Assay Background Reduction: Lowers nonspecific adsorption and hydrophobic interactions in diverse plate-based binding assays.
