YPD Agar with Kanamycin-25 is a rich yeast medium for cultivation of Saccharomyces and other yeasts. The presence of 2% glucose supports fermentative growth, and 25 µg/mL kanamycin suppresses kanamycin-sensitive bacterial contaminants while allowing maintenance of kanamycin-resistant strains such as E. coli bearing kanamycin resistance markers.

  • 1.0% Yeast Extract
  • 2.0% Peptone
  • 2.0% Glucose
  • 2.0% Agar
  • 25µg/mL Kanamycin
  • Yeast Cultivation and Colony Isolation: Rich medium for growing and isolating yeast colonies (e.g., Saccharomyces cerevisiae) while suppressing bacterial contaminants such as Escherichia coli, Pseudomonas aeruginosa, Bacillus subtilis, and Staphylococcus aureus via 25 µg/mL kanamycin.

  • Strain Maintenance and Stocking: Routine passaging, streaking, and preservation of yeast strains with reduced risk of bacterial overgrowth.

  • Colony Purification: Restreaking mixed cultures to obtain pure yeast isolates by suppressing co-occurring bacteria.
