Affinity Equil and Wash Buffer 1, DNase/RNase/Endotoxin Tested, is a ready-to-use buffer for equilibrating and washing affinity chromatography resins and columns. It maintains defined ionic strength and pH during column conditioning, sample application, and wash steps, supporting removal of unbound components prior to elution.

Each lot is tested for DNase, RNase, and endotoxin; endotoxin assessment is performed against E. coli endotoxin standards. The buffer is supplied as an aqueous solution for direct use in affinity purification workflows.

  • Affinity Equil and Wash Buffer 1
  • Affinity Chromatography (His-tagged Proteins): Equilibrates and washes Ni2+/Co2+-charged IMAC resins to remove non-specifically bound proteins prior to elution.

  • FPLC/Chromatography System Prep: Used to condition and wash affinity columns on FPLC systems before sample loading and between runs.

  • Magnetic Beads and Spin Columns: Serves as the equilibration and wash buffer for His-tag capture on magnetic beads or spin columns.

  • Bacterial Lysate Clarifications: Optimizes wash steps during purification from Escherichia coli and other microbial expression systems to reduce contaminants.

  • Low-Endotoxin Workflows: Suitable for endotoxin-sensitive applications, helping maintain low endotoxin burden in downstream cell-based assays.

  • Nuclease-Sensitive Applications: DNase/RNase-tested and sterile buffer supports workflows involving nucleic acids or nucleic acid–binding proteins without introducing nucleases.

  • Sample Preparation for Downstream Analyses: Provides consistent conditions prior to elution for proteomics, enzymatic assays, and structural studies.

  • General Affinity Resin Maintenance: Used for routine column equilibration and intermediate washing to enhance reproducibility and resin longevity.
