YES Agar Plates are a rich agar medium formulated with yeast extract and glucose, supplemented with adenine, histidine, leucine, uracil, and lysine. The medium is solidified with agar to provide a stable surface for colony formation.

Intended for the cultivation and maintenance of yeast strains, including those requiring supplementation with adenine, histidine, leucine, uracil, or lysine. The formulation facilitates growth on a solid medium for colony isolation and routine propagation.

  • 0.5% Yeast Extract
  • 3.0% Glucose
  • 225µg/mL Adenine
  • 225µg/mL Histidine
  • 225µg/mL Leucine
  • 225µg/mL Uracil
  • 225µg/mL Lysine
  • 2.0% Agar
  • Routine Yeast Cultivation and Colony Isolation: Rich, nonselective growth of Schizosaccharomyces pombe on 2.0% agar with 0.5% Yeast Extract and 3.0% Glucose for streaking, colony picking, and culture maintenance.

  • Maintenance of Auxotrophic Strains: Supports strains requiring 225µg/mL Adenine, Histidine, Leucine, Uracil, and Lysine, enabling healthy propagation without selection.

  • CFU Enumeration and Plating Efficiency: Quantifies viability and recovery after stress or treatments using a consistent rich medium formulation.

  • Replica Plating and Strain Purification: Nonselective baseline plating prior to transfer onto selective or differential media for genotype verification.

  • Spot Dilution and Drug Sensitivity Assays: Assess growth differences and compound responses on 3.0% glucose-rich plates for comparative phenotyping.

  • Post-Transformation or Mating Recovery: Allows recovery and outgrowth on supplemented rich medium before applying selection.

  • Temperature-Sensitive and Morphology Screening: Evaluate ts phenotypes and colony morphology on stable 2.0% agar surfaces.
