X-Gal (5-bromo-4-chloro-3-indolyl β-D-galactopyranoside) is a chromogenic substrate for β-galactosidase used for blue/white screening of recombinant E. coli in lacZ α-complementation systems. Enzymatic hydrolysis by β-galactosidase produces an insoluble blue indigo dye, allowing visual differentiation of colonies on selective media.

In practice, X-Gal is incorporated into agar plates, commonly at 20–40 µg/mL, together with an inducer such as IPTG and an antibiotic appropriate for the plasmid backbone, to identify plasmid-bearing transformants in E. coli. It is also used in plaque assays and filter-lift assays where β-galactosidase activity indicates the presence of functional lacZ.
