Tryptic Soy Agar is a general-purpose solid medium formulated for the cultivation, maintenance, and enumeration of a broad range of nonfastidious microorganisms from clinical, food, environmental, and industrial sources. Casein and soybean peptones supply peptides, amino acids, and nitrogen; sodium chloride helps maintain osmotic balance; agar provides a firm surface for colony development.

Representative organisms that can be cultivated include Escherichia coli, Staphylococcus aureus, Pseudomonas aeruginosa, and Bacillus subtilis, among others.

  • 1.5% Tryptone
  • 0.5% Soytone
  • 0.5% Sodium Chloride
  • 1.5% Agar
  • General Microbial Cultivation: Supports robust growth of a wide range of non-fastidious organisms, including Escherichia coli, Staphylococcus aureus, and Bacillus subtilis.

  • Environmental Monitoring: Used for surface and air sampling to detect microbial contamination in cleanrooms and production areas.

  • Clinical Microbiology: Serves as a primary isolation medium for routine culture of specimens to recover organisms such as Pseudomonas aeruginosa and Enterococcus faecalis.

  • Food and Beverage Testing: Employed for total aerobic plate counts and recovery of stressed organisms like Listeria monocytogenes and Salmonella enterica.

  • Quality Control (QC) of Sterility: Used as a growth promotion and sterility test medium to verify microbial detection across diverse species.

  • Colony Morphology and Enumeration: Facilitates colony isolation, morphology assessment, and colony-forming unit (CFU) counting.

  • Maintenance of Stock Cultures: Supports short-term maintenance and subculturing of laboratory strains for routine use.

  • Antimicrobial Screening (Non-selective Base): Provides a non-selective agar base for preliminary antimicrobial efficacy checks and zone observations.

  • Bioburden Testing in Manufacturing: Utilized to assess microbial load in raw materials, intermediates, and finished products.

  • Education and Training: Common teaching medium for basic microbiology techniques and demonstration of bacterial growth.
