Tryptic Soy Agar Plates with 5% Sheep’s Blood are enriched agar plates prepared from a casein–soy peptone base supplemented with defibrinated sheep blood to support the cultivation of a broad range of bacteria, including fastidious species.

The intact erythrocytes enable observation of hemolytic reactions (alpha, beta, gamma) produced by organisms such as Streptococcus spp. and Staphylococcus spp., providing colony morphology and hemolysis patterns on a single medium.

  • 1.5% Tryptone
  • 0.5% Sodium chloride
  • 0.5% Soytone
  • 1.5% Agar
  • 5.0% Sheep's blood (defibrinated)
  • Isolation and Cultivation of Clinical Bacteria: General-purpose growth of aerobic and facultative organisms from clinical specimens, including Streptococcus, Staphylococcus, and Enterococcus.

  • Hemolysis Differentiation: Visual assessment of alpha, beta, and gamma hemolysis for identification of species such as Streptococcus pyogenes (beta) and Streptococcus pneumoniae (alpha).

  • Colony Morphology Assessment: Evaluation of colony size, color, odor, and pigment production for organisms like Staphylococcus aureus and Pseudomonas aeruginosa.

  • Primary Plating in Clinical Microbiology: Initial recovery of potential pathogens from throat, wound, blood, and respiratory samples, including detection of Streptococcus pyogenes.

  • Satellite Test for Fastidious Organisms: Detection of Haemophilus influenzae via satellitism around Staphylococcus aureus streaks.

  • Veterinary and Food Microbiology Screening: Recovery and observation of hemolytic organisms relevant to animal health and food safety, such as Listeria monocytogenes.

  • Antimicrobial Screening (Where Applicable): Qualitative assessment of inhibition around agents on blood-supplemented surfaces for fastidious bacteria when standardized methods permit.

  • Quality Control of Hemolytic Phenotypes: Verification of expected hemolysis patterns using reference strains of Staphylococcus aureus, Streptococcus pyogenes, and related species.
